Chuzhou Mosque stands in the eastern part of the historic Sipailou ('Four Archways') quarter of old Chuzhou, with a history stretching back more than 400 years. Across the centuries it has remained the religious and social center of the city's small Hui Muslim community, which settled in this old commercial district around the late Ming and early Qing period.

Several local buses stop near the Sipailou area in old downtown Chuzhou.
There is no significant Arab or large Muslim quarter nearby in Chuzhou itself; the nearest major mosque is Jingjue Mosque in neighboring Nanjing, about an hour's drive away.
The mosque sits within the historic Sipailou quarter in Chuzhou's own downtown, a few minutes' walk from the central square.
